To identify the miRNA expression profile of Herpes Simplex Keratitis(HSK) in C57BL/6 mice
Mice cornea was infected with herpes simplex virus-1(HSV-1) and then microRNA microarray technique was performed to identify differentially-expressed miRNAs. Some miRNAs were consquently verified by real-time quantitative PCR (qRT-PCR). Functions and locations of theses miRNAs were predicted by databases.
Upregulation of some microRNAs was identified in HSK cornea including mmu-miR-223-3p, mmu-miR-155-5p, mmu-miR-21-3p, mmu-miR-150-5p, mmu-miR-18a-5p, mmu-miR-3096b-5p, mmu-miR-146a-5p and mmu-miR-342-3p. While, downregulation of mmu-miR-434-3p, mmu-miR-124-3p, mmu-miR-204-5p, mmu-miR-133b-3p, mmu-miR-206-3p and mmu-miR-133a-3p was found in HSK. Moreover, the differential expression of mmu-miR-204-5p, mmu-miR-206-3p, mmu-miR-155-5p, mmu-miR-133a-3p and mmu-miR-150-5p were identified by qRT-PCR in HSK cornea tissues. Their putative target genes were predicted using 2 online software programs
Some new miRNAs are found in HSK, which might play significant roles in regulating in pathological process.
