Abstract

Purpose: The aim of this study was to evaluate the adverse effects of particulate matter on the ocular surface. Titanium dioxide (TiO2) nanoparticles, one of engineered nanoparticles were used for this study

Methods: Forty eyes of 40 New Zealand white rabbits were used. TiO2-1D group included 10 rabbits which received instillation of 0.1㎖ TiO2 (10㎍/100㎕) in the right eye once a day for 1day. TiO2-4D group included 10 rabbits which received instillation of 0.1㎖ TiO2 in the right eye once a day for 4 days. The remaining 20 rabbits were allocated to normal control 1 (n=10) and 2 (n=10) for each TiO2 group. Lactic dehydrogenase (LDH) and MUC5AC concentrations in tear were measured and densities of conjunctival goblet cell were calculated through impression cytology 24 h after final instillation.

Results: The median tear LDH was 1.39 opitcal density(OD) in the TiO2-1D group and 0.58 OD in the TiO2-4D group, which were significantly higher than those of each control group (0.57 and 0.29 OD, respectively) (P=0.016, P=0.008, respectively). However, an increase in tear levels of MUC5AC was only observed in the TiO2-1D group, but not in the TiO2-4D group, compared to control groups. The median conjunctival goblet cell density in the TiO2-1D group, 13.6%, and the TiO2-4D group, 11.8%, was significantly lower than that of each control group, 23.6% and 22.1%, respectively (P=0.008 and P=0.008, respectively).

Conclusions: Exposure to TiO2 nanoparticles increased LDH level in tear and decreased the conjunctival goblet cell density in rabbit eyes. Although exposure to TiO2 nanoparticles increased MUC5AC level in tear at first, MUC5AC level was decreased to normal level after repetitive TiO2 application.
