Abstract

Purpose: : To asses the intrasession reproducibility of peripapillary retinal nerve fiber layer (RNFL) thickness measurements by the Stratus Optical Coherence Tomograph (Stratus) and OTI spectral OCT/SLO (Opko Health Inc, AMEX: OPK, Miami, FL) OCT systems in glaucomatous eyes

Methods: : Global and sectorial RNFL thickness was obtained and analyzed with both OTI and Stratus OCT systems. Bland and Altman plots were constructed to assess intrasession repeatability. Precision (repeatability coefficient) was defined as mean difference ±1.96 the standard deviation (SD) of the differences. Coefficients of variation (CV) were also calculated for each RNFL quadrant (temporal, superior, nasal, inferior) for the two different OCT systems. Wilcoxon sign-rank test Pair-wise was used to analyze CV differences between instruments and a p<0.05 was considered statistically significant. Comparisons of each of RNFL quadrant CV between the 2 different OCT systems were performed with paired t-test.

Results: : Twenty-five eyes of 25 glaucomatous subjects (mean age 66.8 ±7.9 years) were considered. The mean difference of test-retest measurements for the Stratus OCT for the inferior-, nasal-, superior- and temporal-quadrant RNFL thickness was 0.2 µm (12.23/-11.83, upper and lower SD level respectively), -0.8 µm (17.26/-9.16), 3,9 µm (31.7/-13.4), 1.13 µm (15.4/13.16) and for the OTI OCT 4.06 µm (21.76/-13.6), 4.76 µm (25.26/-15.73), 1.8 µm (24.2/-20,6), 0.06 µm (13.6/-13.46) respectively. CV of mean RNFL thickness measurements for the inferior-, nasal-, superior- and temporal-quadrant was: 4.4%, 9.7%, 5.8%, 7.8% respectively for the Stratus OCT and 5.2%, 7%, 4.9%, 5.9% respectively for the OTI OCT. The difference between CV of different RNFL quadrants between the two OCT systems was not statistically significant (p<0.05%).

Conclusions: : RNFL thickness measurements is highly and similarly reproducible in glaucomatous eyes both with Stratus and OTI spectral OCT/SLO systems.
