Abstract

Abstract: : Objective:To compare single–doses of ophthalmic pemirolast potassium 0.1% (P) and olopatadine hydrochloride 0.1% (O) in the prevention and treatment of ocular itching in cat sensitive individuals. Methods: We conducted two double–masked, randomized "cat room" studies in subjects with documented cat–dander ocular allergy. Study 1 evaluated the effects of a single–dose 20 minutes prior to allergen exposure and study 2 assessed a single–dose 30 minutes after entry and an itching score of >2+ (mild or greater) response to the "cat room." A randomized, masked dose of each agent was administered topically to each eye. Study 1 subjects were assessed at 5, 15, 30, 45, and 60 minutes. Study 2 patients were evaluated at 15, 30, 45, and 60 minutes for >2+ itching. Ocular signs of allergy were evaluated with a slit lamp examination and the mean itching score at each time point by questionnaire. Treatment scores were compared statistically utilizing an independent sample t–test. Forced expiratory volume at 1 second (FEV1) and adverse events were followed for safety. Results: 50 subjects were evaluated in each study. In study 1, mean itching scores at 5, 15, 30, 45, and 60 minutes were lower in eyes pretreated with P than those with O (5 min, P = .16, O = .24; 15 min P = .26, O = .44; 30 min P = .36, O = .56; 45 min P = .34, O = .56; 60 min P = .32, O = .58). No statistical significance was shown between the two groups (p>=0.10). In study 2, the mean itching scores at 15, 30, 45, and 60 minutes were roughly equivalent between groups (15 min P = 1.16, O = 1.13; 30 min P = 1.16, O = .9; 45 min P = .70, O = .68; 60 min P = .51, O = .53) with no statistical significance shown between the two groups (p>=0.12). Slit lamp examination did not reveal clinically significant signs of ocular allergy in eyes treated with O and eyes treated with P. Both medications were well tolerated in each study and no adverse events were reported. Conclusion: When given as single–doses, P and O appear comparable in the prevention of ocular itching when administered 20 minutes prior to allergen challenge in cat sensitive individuals. Additionally, both agents are effective in controlling itching up to 60 minutes during allergen exposure.
