Purpose: Natriuretic peptides can modulate ocular hydrodynamics and certain ocular hypotensive drugs elevate levels of ANP in aqueous humor. This study is to examine the effects of ANP, BNP, and CNP on: 1) expression of natriuretic peptide receptor A-type (NPR-A), B-type (NPR-B) and C-type (NPR-C) and 2) cGMP and cAMP levels in human nonpigmented ciliary epithelial (HNPE) cells, trabecular meshwork (HTM) cells and retinal precursor (HRP) cells. Methods: Expression of NPR-A, NPR-B and NPR-C receptors was measured by RT-PCR analysis. Levels of cGMP and cAMP were determined by enzyme immunoassay. Results: NPR-A (491bp), NPR-B (768bp) and NPR-C (362bp) were expressed in HRP cells. Both NPR-A and NPR-B mRNAs were shown in HNPE cells. Furthermore, HTM cells demonstrated NPR-A and NPR-B mRNAs more prominently after pretreatment with ANP (10µM), BNP (10µM) and CNP (0.1, 1, 10µM) for 24 hr. Functional studies, HTM cells, demonstrated that in CNP enhanced basal cGMP levels by 25% and inhibited forskolin-stimulated cAMP accumulation by 50%. Conclusion: NPR-A, NPR-B and/or NPR-C were expressed in HRP cells and HNPE cells. NPR mRNAs were induced by pretreatment with ANP, BNP and CNP. Functional evidence indicated that alteration of cGMP/cAMP by CNP is mediated by activation of NPR suggesting that NP system may be the part of mechanisms of regulation of aqueous humor dynamics and neuroprotection in the eye.
