Tranilast release from the polymer laminate was studied in phosphate-buffered saline (PBS). A polymer film sample (1 cm × 1 cm) was placed in 23 mL PBS (pH 7.4) containing 0.1% sodium azide as a preservative, and magnetically stirred in a circulating water bath 37°C. At predetermined times (0, 0.17, 0.25, 0.33, 0.42, 1, 2, 3, 5, 6, 7, 10, 15, 17, 21, 27, and 30 days), 400-μL aliquots of the medium were collected, replaced with an equal volume of fresh PBS, and filtered through a 0.45-μm membrane filter. Ten microliters of this solution was directly injected to the HPLC for tranilast quantification. In vitro release experiments were repeated three times to obtain a release profile of tranilast.
