In Vivo Imaging of hESC-RPE Implanted in the RCS Rat Using InfraRed Reflectance

Abstract

Purpose: To investigate if the Infrared Reflectance <IR> imaging modality was able to detect the in-vivo presence of RPE cells derived from embryonic human stem-cells (hESC-RPE>implanted into the subretinal space of the Royal College of Surgeon <RCS> rat

Methods: Monthly IR images were obtained from RCS rats implanted with hESC-RPE seeded on a scaffold made of parylene<n=14> or the parylene scaffold alone <n=4>. Two independent masked investigators graded the images. Histology and immunohistochemistry were performed at different time point P150, P210 and P270>

Results: IR images reveled that an average of 20.53‰ of the parylene membrane area was covered by hESC RPE. RPE65 and TRA 185 confirmed the presence of human specific RPE cells in those animals. No areas corresponding to cells were found in the group implanted with membrane without cells. Agreement between the graders was high<r=0.89-0.92>

Conclusions: The infrared mode was suitable to detect the presence, location and distribution of hESC RPE seeded on a parylene scaffold membrane implanted into the subretina space of the RCS rat.
