Protein was extracted from gel samples following TRIzol manufacturer's instructions. Equal amounts of protein were loaded onto and run on 12% precast polyacrylamide mini-gels (Thermo Fisher Scientific, Northumberland, UK) and then transferred, using a semi-dry transfer system (Bio-Rad, Hercules, CA) onto polyvinylidene fluoride (PVDF) membranes (Thermo Fisher Scientific). Membranes were blocked in 3% reduced-fat milk powder in 0.05% Tween-20 in phosphate buffered saline (TBS-T) followed by overnight incubation at 4°C in primary antibody (MMP1; 1:500, Abcam, Cambridge, UK or Rac1; 1:4000, BD Transduction Laboratories, Oxford, UK). Membranes were washed 3 times for 10 minutes in TBS-T, incubated in secondary-labeled antibody (IRDye 680, 1:5000; LI-COR, Cambridge, UK) for 2 hours at room temperature, washed, and scanned on an Odyssey IR Imager (LI-COR).
