Abstract

Purpose: To evaluate retinal pigment epithelial (RPE) morphology in eyes with neovascular age-related macular degeneration (AMD) using wide-field polarization sensitive optical coherence tomography (PS-OCT) in correlation with functional microperimetry measurements.

Methods: Ten eyes of 10 patients with neovascular AMD were imaged by a novel wide-field PS-OCT system (imaging an area of 40x40°) capable of specifically detecting the RPE based on its inherent tissue-specific contrast. In addition the device visualizes outer retinal layers based on intensity. En face maps of RPE morphology obtained by PS-OCT were correlated to microperimetry findings.

Results: RPE atrophy was detected reliably in all eyes by PS-OCT. Overlays of PS-OCT maps and microperimetry images showed reductions of retinal sensitivity within regions revealing atrophic RPE. Retinal fibrosis and damage of photoreceptor integrity was further associated with reduced retinal sensitivity.

Conclusions: Wide-field PS-OCT specifically detects the RPE and associated lesions in eyes with neovascular AMD allowing for the correlation of RPE integrity and function. RPE atrophy corresponds to sensitivity loss in neovascular AMD.
