Abstract
Purpose:
To learn whether decellularized extracellar matrixs might modulate the inflammation tear secretion of cultured rabbit lacrimal acinar cells induced by il-1
Methods:
An in vitro dry eye model was produced by adding il-1b to the culture medium of primary rabbit lacrimal acinar cells. Decellarized extracellular matrixs of porcine lung, cartilage, cornea and lymph node were applied to the model resperately for 48 hours. Inflammatory cytokins expression, including il-8, il-17, tnf and mmp-9, and tear proteins expression, including AOP5, EPCAM, LTF, CDH3 and OCLN were evaluated by RT-PCR. Tear proteins were induced by carbachol and detected by β-N-Acetylglucosaminidase Assay.
Results:
In cultured rabbit lacrimal acinar cells, none of the decellularized extracellular matrixs affected the cell growth. The decellularized extracellular matrixs upregulated gene expression of tear proteins but did not modulate il-1 induced inflammation. Carbachol only elevated tear proteins secretion in the decellularized extracellular matrix of porcine lymph node treated group.
Conclusions:
Decellularized extracellular matrix might be a useful supplementary treatment agent agaist dry eye disease where the immunoinflammation is well controled.
