Abstract
Purpose: :
To investigate the effect of Combretastatin A4 Phosphate(CA4P) on proliferation and migration of lens epithelial cells.
Methods: :
Human lens epithelial cells line SRA01/04 was treated with or without CA4P for 15 mins.The antiproliferation effect was assessed by WST - 1 and the cell cycle stage was detected by Cytometry under PI staining. The ability of endothelial cells to migrate and form tubular structures was detected by a wound assay. Programmed cell death was detected by Annexin V - FITC/PI. Cytoskeleton protein (alpha microtubules protein, F-actin) and cell morphology were detected by immunofluorescence.
Results: :
CA4P induced time-dependent antiproliferative activities against lens epithelial cells, and CA4P caused a G2/M arrest. The in vitro wound assay and tube formation assay indicated that CA4P significantly inhibited migration of lens epithelial cells and tube formation. Apoptosis was potently induced and abnormal cell morphology was observed in CA4P-treated epithelial cells.
Conclusions: :
CA4P inhibits proliferation and migration of lens epithelial cells. This can be a novel prophylactic for posterior capsular opacification.
