Abstract

Purpose: : This study was undertaken to investigate the NK cell-associated Ag expression in retinoblastoma (RBL) animal model.

Methods: : Xenoplantation mouse specimens primarily enucleated after 4 weeks of inoculation of two different RB cell lines those are Y79 and SNUOT-Rb1 were stained to detect NK cell-associated Ag, including HLA-G, MICA, MICA/MICB, and ULBP2. Using immunofluorescence, the expression of these proteins, and their relative locations were also analyzed.

Results: : In both xenoplantation tumor model injected with immortalized retinoblastoma cell lines showed the cytoplasmic staining of HLA-G and MICA/MICB. Interestingly, these proteins were strongly expressed along the periphery of the tumor bed. Immunofluorescence staining also demonstrated stronger expression of HLA-G and MICA/MICB in proliferating tumor cells located in the periphery of the tumor bed.

Conclusions: : Our data revealed that HLA-G and MICA/MICB are strongly expressed in the proliferating retinoblastoma tumor cells that located in the periphery of the tumor bed. Our data also suggests that NK cell might be a promising tool for targeted cell therapy against retinoblastoma.
