Abstract
 
Purpose:
 

To evaluate the effects of PDGF-BB and bFGF on proliferationand cytokine production of orbital fibroblast

 
Methods:
 

Orbital fibroblasts were cultured(5 from eyes of thyroid-associatedophthalmopathy, 5 from normal eyes) and classified into 3 groups: (1) Control group, (2) PDGF-BB stimulation group, (3) bFGFstimulation group. Cytokine production(IL-1ß, IL-2, IL-6,IL-8, RANTES) of each group was measured. Proliferation of orbitalfibroblast was measured in each group using a thymidine analogEdU, which can be incorporated into DNA during active DNA synthesis

 
Results:
 

Only IL-8 showed a borderline significant increase in group2 than group 1(p=0.085). The other cytokines showed no significantdifference. IL-6 and IL-8 production was increased in group3 but it was not statistically significant. Proliferation oforbital fibroblasts was increased from twice to quadruple ingroup 2, but there was no difference in group 1 and 3

 
Conclusions:
 

PDGF-BB induced orbital fibroblast proliferation and increasedproduction of IL-8. Further study for the stimulating factorsof orbital fibroblasts with larger sample size is needed andwe may expect the fundamental treatment of thyroid-associatedophthalmopathy
