Murine corneal lymphatic vasculature has been shown to be influenced by the genetic background: C57BL/6 mice showed significantly more resting limbal vasculature compared to Balb/c and FVB mice. To gain insight into the molecular characteristics between the strains we analyzed a panel of angiogenesis and endothelial cell biology related genes in the untreated cornea of C57BL/6, Balb/c and FVB mice using PCR-based gene expression profiling.
2mm punches of the center of untreated murine corneas (C57BL/6, Balb/c and FVB mice) were homogenized using the PRECELLYS® 24 tissue homogenizer. Following RNA extraction cDNA synthesis was carried out with SuperScriptTM III First-Strand Synthesis SuperMix. The cDNA was used for real-time PCR analyses with the Angiogenesis and Endothelial Cell Biology RT² ProfilerTM PCR Array (Superarray). The expression of 84 pathway specific genes was compared between the three different mouse strains.
Differential gene expression analyses showed a down-regulation of molecules with cytokine activity (e.g.CXCL1, CXCL2) and adhesion molecules (e.g.THBS1) in the untreated cornea of Balb/c or FVB mice compared to the C57BL/6 mice. Furthermore several molecules were higher expressed in the cornea of Balb/c or FVB mice including proteases like PLAU, PLAT and the endothelial specific receptor tyrosin kinase TEK.
By using angiogenesis-specific and endothelial cell biology-specific analyses several differential regulated genes in the cornea of C57BL/6, Balb/c and FVB mice were identified. Information on the strain specific molecular expression profile of the cornea could contribute to our understanding of interindividual differences in the resting limbal vasculature.
