Abstract

Purpose: : The objective of the present study was to evaluate the effect of aging on the structure and the secretory mechanisms of the LG.

Methods: : Male Wistar rats aged 24 and 2 months were used. Tears secretion and impression cytology of the cornea were compared. The LG were removed and submitted to structural and morphologic comparison, and analysis of the expression of peroxidase, acetylcholine, vitamin E and MDA. RT PCR was used for the analysis of mRNA expression.

Results: : Impression cytology demonstrated increased metaplasia in the cornea of the aging group (p=0.0320). The Schirmer test was similar in both groups (8.8 ± 1.0 mm for the control and 7.3 ± 1.4 mm for the aging group, p=0.5145). The LG of the aging group showed increased MDA (11.0 ± 0.89 mmol/g and 24.6 ± 1.65 mmol/g, p=0.0571) and peroxidase (1.15 ± 0.52 and 1.05 ± 0.09 mU/mg of LG weight, p=0.34) and reduced vitamin E (4.98 ± 0.50 µM and 1.79 ± 0.18 µM, p=0.0159). The expression of Rab 27b and Rab 3b mRNA was confirmed and decreased with aging (p< 0.05).
