Abstract

Purpose: : to evaluate nuclear NF-kB translocation in minor salivary glands (mSG) of human primary Sjögren Syndrome (pSS) and lacrimal gland of NOD mice.

Methods: : Biopsies of lip's mSG were made in 21 female patients with diagnosis of pSS arised from the Rheumatology Service of Rivadavia Hospital and lacrimal gland (LG) of 10 male NOD mice (12 week old). Human and mice glands were staining with H&E and PAS and immunostaining for NF-kB. Human specimens were classified according to the Chisholm and Masson score.

Results: : The biopsies from patients with pSS (H&E staining) showed lymphoplasmocitic infiltrates, forming periacini and periductal focuses which number depends on the stage of the disease. In stages III and IV it's observed acini destruction and, in some cases, fibrosis. In the biopsies with sialadenitis diagnosis we observe interstitially-dispersed lymphoplasmocitic elements and also polimorphonuclear neutrophils. The lip's mSG biopsies of patients with clinical-serological diagnosis of pSS showed nuclear translocation of NF-kB in the lymphocytes of focal infiltrations and in the acini epithelium adjacent to the infiltrates. In acini and ductal structures far from the infitrates we didn't observe nuclear translocation. However, in patients with sialadenitis it was observed mononuclear leukocytes interstially. Patients with sicca syndrome associated to other causes didn't show nuclear translocation of NF-kB factor neither in acini nor in ductus. LG of NOD mice showed nuclear NF-kB translocation in lymphocytes and in acini and ductal structures adjacent to the infiltrates.

Conclusions: : These results let us infer the importance of epithelium-lymphocyte interaction on the activation of factor NF-kB of human pSS and NOD mice
