Abstract
Purpose: :
Toll-like receptor 2 (TLR2) is a member of the TLR family, which plays a central role in the innate immune response to a wide variety of microorganisms, including bacterial, fungal, protozoal and viral molecules due to interaction with TLR1 and TLR6. Polymorphisms for TLR2 gene (Arg677Trp, Arg753Gln) are associated with high risk infection of Mycobacterium sp, Candida albicans, Herpes Simple Virus Type 2 and others. Recent studies have showed that TLR2 gene polymorphisms withing the TIR domain (amino acids 643-784) are critical to TLR dimerization and signaling. The present study aimed to investigate the relationship between TLR2 gene polymorphisms and adenoviral ocular infection in a Mexican population.
Methods: :
Thirty three samples from patients with sporadic and epidemic adenoviral conjunctivitis, and ten samples from not infected subjects as controls, were studied. Genomic DNA was extracted. A region TLR2 gene which codes the cytosolic part of this receptor was amplified by PCR and analyzed by direct sequencing.
Results: :
We did not found heterozygous for the TLR2 Arg677Trp and Arg753Gln in all subjects. However, we found the TLR2 Phe707Phe in five patients that is a heterozygous frequency of 15%. Control samples do not showed any polymorphisms reported in patients with adenoviral ocular infections.
Conclusions: :
These results showed that the polymorphisms Arg677Trp and Arg753Gln are not present in the Mexican population studied, but the Phe707Phe polymorphism that was reported in Hispanic population, could play an important role in the infection risk by adenovirus, however, it is important to include a large number of subjects in the study.
