Abstract

Purpose: : Molecular genetic testing of uveal melanomas (UM)can identify patients who have increased risk for development of metastatic disease. However there is currently no effective systemic therapy for aggressive UM and patients with increase risk for an aggressive disease are only followed up with imaging until the time of metastasis. Identifying systemic treatment or chemoprevention of metastatic UM could improve the prognosis of this malignancy. The anti-neoplastic effect of several spices and natural products is well recognized.

Methods: : Methanolic extracts of several medicinal plants were screened for their anti-neoplastic activity on four UM cell lines (C918, 92.1, MEL270 and OCM3) and 2 non-neoplastic control cell lines (ARP19 and MCR5). Extracts showing selective activity on UM cells were further purified by solvent fractionation and chromatography in an attempt to identify the antineoplastic components.

Results: : Out of 41 different extracts screened, seven showed significant inhibition of cell proliferation of the UM cell lines at or below concentrations of 25 µg/ml. Promising extracts, including Rosmarinus officinalis, Origanum syriacum and Mentha longifolia, showed selective inhibition of UM cell lines compared to normal control cells. Activity-guided fractionation of candidate extracts is underway to identify the active components.Slight variation of the response of UM cells to cell proliferation inhibition by various extracts was observed which could be linked to the genetic alterations in BRAF/GNAQ identified in the tested cell lines.

Conclusions: : Our results indicate the potential antineoplastic effect of several medicinal herbs and spices in UM. The utility of these herbs in the management of high-risk UM patients will need to be further explored. Finally, molecular alterations in the tumor may influence the degree of responsiveness to herbal therapy and such correlation should be further explored.
