To analyze the qualitative and quantitative effects of laser-photocoagulation (LP)-induced ocular hypertension (OHT) in the macroglia and retinal ganglion cells (RCGs) of OHT and contralateral eyes.
Adult albino Swiss mice were divided in two groups: Age-matched control; (G0, n=6): Group 1 (G1, n= 7):designed to determine the effects of OHT in treated and contralateral eyes one week after LP. Retinal whole-mounts were double immunostained with anti-GFAP to test OHT effects on retinal macroglia plus anti-NF-200 and to analyze the presence of degenerated RGCs. The area occupied by astrocytes (AOA) and the number of astrocytes and NF-200(+)RGCs were quantified.
In G0 the astrocytes formed a homogenous plexus on the nerve-fiber and RCG layer of GFAP(+) stellated cells regularly distributed throughout the retina. Astrocytes in OHT eyes had thinner cells bodies and primary processes and fewer secondary processes than in G0. Compared with G0, astrocytes of contralateral eyes were more robust, forming a honeycomb-like network. Astrocyte number did not differ significantly between the study groups. However, the AOA in OHT eyes was reduced compared with contralateral and G0 (p<0.000) and the contralateral eyes had more AOA than G0 (p<0,032). Müller cells were GFAP(+) in G0 and G1 while immunostaining was greater in eyes with OHT. NF-200(+)RGCs appeared only in OHT eyes with a trend for the AOA to decrease and for the RGC number to increase from the center to the periphery (r=-,448).
After one week of OHT the contralateral eye changed in the astrocytes, and thus these fellow eyes should not be used as control. OHT reduced the area of the retina occupied by astrocytes, which seems to be inversely related to the number of NF-200(+)RGC.
