Abstract

Purpose:

To compare and analyze the expression of GRP78 in trabecularmeshwork between eyes with primary open angle glaucoma(POAG)and normal eyes.

Methods:

The human trabecular meshwork cells were cultured from eyeswith POAG and normal ones. After incubated with different irritation--Tunicamycin(TM), Staurosporine(STS) and DMSO for 6,24 and 48hours, immunofluorescence ,laser confocal tmicroscopy were employedto observe the induction function of TM in trabecular meshworkcells’ death. The different expression of GRP78 mRNA wasdetected by RT-PCR.

Results:

The results of immunofluorescence displayed that GRP78 and myocilinwere part co-location. The expression of GRP78 mRNA in eyeswith POAG was reduction compared with normal adults. This differencewill become obviously when treated with TM,otherwise this phenomenondidin’t appear in control groups.

Conclusions:

The expression of GRP78 in trabecular meshwork celles of eyeswith POAG decrease, which implied that the endoplasmic reticulumstress may play an important role in the process of POAG onset.
