The Effects of Fibronectin on Proliferation, Attachment and Migration of Human Lens Epithelial Cell

Abstract

Purpose: : To investigate the effects of fibronectin on the proliferation, attachment and migration of human lens epithelial cell line HLE-B3 and the expression of its receptor integrin α5.

Methods: : Human lens epithelial cell line HLE-B3 was cultivated and inoculated in plates coated by FN in different concentration.The biologic characteristics of HLE-B3 was observed. BrdU labeling and WST-8 method was used to detect the proliferation and attachment of the cells. Scratch method was used to observe the migration of the cells, the hours of the closure of cell defect domain was recorded. Western-blot method was used to detect the expression of integrin α5 quantitatively of each FN concentration group.

Results: : After coating with FN in different concentration, BrdU labeling staining showed that there was no difference in each FN concentration. As the coating concentration of FN raised, the absorbance of WST-8 method increased gradually in attachment essay. The cell defect domain closure time were shortened gradually in scratch essay. Integrin α5 expression was enhanced as the coating concentration of FN raised in western blot assay.

Conclusions: : FN can significantly promote the attachment and migration of HLE-B3 as well as up-regulating the expression of its receptor integrin α5, but the effect on the proliferation of HLE-B3 was not obvious.
