Abstract

Methods:: Transgenic Nrl-/- mice underwent timed pregnancies. At embryonic day 14 (E14), the left subretinal space was injected with an adeno-associated virus encoding the wildtype nrl gene (AAV. CMV2/5.nrl). Non-injected contralateral eyes served as controls. At postnatal day 45 (P45) electroretinograms (ERG) and immunohistochemical analysis for rod-photoreceptor specific proteins were performed.

Results:: Experimental and control eyes exhibited no significant ERG differences. However, histological sections exhibited rhodopsin and transducin positive photoreceptors cells.

Conclusions:: In utero gene delivery can alter isolated photoreceptor cell differentiation from a cone pathway to a rod pathway evidenced by the expression of rod-specific proteins. Earlier delivery may increase the number of cells with a rod photoreceptor fate. Future visual behavioral analysis will also evaluate rod cell function.<br /
