We sought to identify genetic changes in the retinae of trVEGF029, an animal model that displays pathological changes consistent with non-proliferative and early stages of proliferative diabetic retinopathy.
We used microarray to compare the genetic profile of the posterior eye cups of trVEGF029 to wild-type mice. Real-time PCR was used to confirm the changes in gene expression of a previously uncharacterised gene, NM_177853. The expression was studied between 1.5 and 15 weeks post-natal age of trVEGF029 and wild-type eyes. Multiple tissue expression was also identified by qPCR and immunohistochemistry of eye sections.
We identified a previously uncharacterised gene (NM_177853) upregulated in the posterior eye cups of trVEGF029. NM_177853 expression was shown to increase with age and development of retinal vascular changes, and is upregulated following mVEGF stimulation and hyperglycemia. It is expressed primarily in the retinal pigment epithelium and choroid of the eye, and in areas of the brain, kidney, ovary and embryo.
Here we report the first analysis of NM_177853 a new gene associated with VEGF-induced retinal neovascularisation.
