Comparative Cytotoxicity of Contact Lens Care Products and Contact Lenses Using the USP Direct Contact Test

Abstract

Purpose: : Contact lenses cycled in soft contact lens care products were evaluated for cytotoxicity according to the USP Direct Contact Test.

Methods: : The USP/ISO Direct Contact Test for cellular viability was used to evaluate the interaction of Group 1, Group IV and HiDk SiHy contact lenses with 3 soft contact lens care products (SCLC). Group I– low water/nonionic(CIBA Soft), Group IV– high water/ionic (Focus Monthly) and HiDk SiHy contact lenses (NIGHT & DAYTM and O2OPTIXTM [CIBA Vision], PureVisionTM [Bausch and Lomb], and ACUVUE® ADVANCETM [Vistakon] were cycled for 1 and 7 days prior to testing. The lens care products used were: Clear CareTM and AQuifyTM MPS(aka SOLOcare®AQUA) [CIBA Vision] and ReNu MPS® with Moisture LocTM [Bausch and Lomb]. Following an incubation period, L929 cells were exposed to 1 and 7 day SCLC cycled contact lenses for 24 hours and cellular viability was evaluated by Trypan Blue, an exclusionary dye.

Results: : Trypan blue cell viability was significantly different between the 3 SCLC solutions and the various contact lenses. Direct contact analysis demonstrated that Group I, Group IV and all HiDk SiHy lenses cycled for 1 and 7 days with Clear CareTM and AQuifyTM MPS were noncytotoxic to L929 cells. Group I and Group IV lenses cycled for 1 day with ReNu MPS with Moisture Loc were found to be cytotoxic whereas all HiDk SiHy lenses cycled for 1 day with ReNu MPS with Moisture Loc were noncytotoxic. Group I, Group IV and all HiDk SiHy lenses except for PureVisionTM cycled for 7 days with ReNu MPS with Moisture Loc were cytotoxic.

Conclusions: : Direct contact results of 1 and 7 day cycled lens studies demonstrate the biocompatibility of Clear CareTM and AQuifyTM MPS with Group 1, Group IV, and all HiDk SiHy lenses tested with L929 cells. All lenses cycled for 7 days with ReNu MPS with Moisture Loc except for PureVision do not exhibit biocompatibility with L929 cells according to the USP Direct Contact Test.
