Abstract

Purpose: : To determine the expression levels of VEGF and PEDF after moderate cryotherapy and photocoagulation in eyes porcine model.

Methods: : 9 eyes of nine pigs were included. Eyes were separated in 3 groups. Group 1 (n=3) treated with moderate submacular cryotherapy (mild whitening effect); group 2 with argon laser photocoagulation (n=3); and control group (n=3). Eyes were enucleated every week during 3 weeks. Expression levels of VEGF and PEDF in choroids and retinal were quantified by RT–PCR using specific primers for produce fragments 191 and 225 bp respectively. Proteins in the tissue and vitreous were determined inmunohistochemical and western blot using polyclonal antibodies IgG anti–VEGF and anti–PEDF (Santa Cruz ).

Results: : the levels mRNA of VEGF and PEDF were quantified by RT–PCR. VEGF decreased after mild cryotherapy and laser photocoagulation, difference was statistically significant. The protein levels of VEGF in tissue and vitreous showed concordance with above results. Actually PEDF analyses is processing.

Conclusions: : Mild cryotherapy and laser photocoagulation applied to retina decreased VEGF levels in retina of pig eyes.
