Abstract

Abstract: : Purpose: We previously reported that genes for immediate early responsible transcription factors and cytokines were highly up–regulated in iris–ciliary body (ICB) at very early stage of endotoxin–induced uveitis (EIU) by using an oligonucleotide microarray system. The purpose of this study is to investigate some of stress genes up–regulated in ICB with EIU and to discuss possible roles of the genes in EIU. Methods: EIU was induced by a footpad injection of 200µg lipopolysaccharide (LPS) in male Lewis rats. ICB was collected at 0, 2, 6, 12, and 24 hours after LPS injection. Total–RNA was isolated and real–time RT–PCR was performed. Aqueous humor (AqH) was collected at 0, 3, 6, 12, 24 and 48 hours after LPS injection, total superoxide dismutases (SOD) activity in these samples was measured by using SOD assay kit (Cayman Chemical Company). Expression of heat shock protein 70 (HSP 70) was also evaluated through immunohistchemistry on cryosections of enucleated eyes with anti–HSP 70 antibody (Santa Cruz Biotechnology). Results: The mRNA expression of SOD 2 and HSP 70 increased at 6 hours after LPS injection, 32 and 27 fold over control respectively, and then decreased. A significant increase in total SOD activities was found in AqH after 12, 24, 48 hours after LPS injection as compared with normal control. HSP 70 immunoreactivity was found in ICB cells at 24 hours after LPS injection. Conclusions: The results of this study demonstrated that there were protein expressions of SOD and HSP 70 as well as genes in the ICB with EIU. SOD is well–known as a free radical scavenger that is the first line of defense against cytotoxic superoxide. HSP 70 is the component of a molecular chaperone complex that can assist the folding of particular proteins. SOD and HSP 70 may play a protective role in EIU.
