Abstract

Abstract: : Purpose: To determine the effect of clusterin on retinal endothelial cell in ischemic retina Methods: At 6, 12, 24, 48 hours after permanent ipsilateral carotid artery ligation of the Sprague–Dawley rats, immunohistochemistry and western blotting for clusterin, PECAM, and ZO–1, and TUNEL were performed in the retina. MTT assay was done in hydrogen peroxide – treated cells with clusterin overexpression. Results: In the ipsilateral retina, clusterin expression was progressively enhanced with increasing time. Clusterin was prominently expressed in inner retina. In contrast, PECAM and ZO–1 were inversely decreased. TUNEL was evident at 24 hours. Clusterin overexpression did not upregulate cell death. Conclusions: Ischemia affects the expression of clusterin in the retina. Clusterin expression may reflect an ischemia–induced damage and/or may protect retinal endothelial cell from ischemic injury.
