Abstract

Abstract: : Purpose: To characterize behavioral and electrophysiological changes in mice after intravenous (iv) administration of sodium iodate and to correlate them to the retinal pigment epithelium (RPE) loss occurred. Methods: 1% sterile sodium iodate (NaIO3) was injected iv into 4–6 week old C57BL/6 mice with a single injection of 50 mg/kg. After 1, 3, 7, 14, 21, and 28 days a cued water maze test was performed followed by ERG recording. Additionally, autofluorescence of whole eye flat mounts was detected and H&E staining of paraffin sections of the enucleated eyes were performed to observe the overall changes in the retinal anatomy. Results: One day after the NaIO3 treatment no changes in behavior and retinal anatomy were detectable, but the ERG already showed an altered b–wave. On day 3, the ERG remained diminished, the cued water maze test became abnormal and patchy RPE cell loss was observed on microscopy. On day 7, RPE loss was more extensive, the outer nuclear layer (ONL) thickness was reduced. On day 14, the number of nuclei in the ONL was reduced as well and the ERG was extinguished. Conclusions: We observed a direct correlation between anatomical cell loss, decreased visual function and decreased electrophysiological function in the rodent after sodium iodate injection. Thus, this model is quite suitable for the study of the functional effect of retinal or stem cell transplantation into an altered retinal environment.
