Abstract

Abstract: : Purpose: Kir7.1 is an inwardly rectifying K+ channel subunit with unique functional properties that plays a critical role in K+ transport and Na+–K+ ATPase function in epithelia, most notably the retinal pigment epithelium and choroid plexus. The aim of this study was to determine whether Kir7.1 might also be expressed in association with Na+–K+ ATPase in the ciliary epithelium, the tissue responsible for aqueous humor secretion. Methods: Ciliary bodies were isolated from freshly procured porcine eyes, fixed in 4% paraformaldehyde and cryo–preserved. Immunohistochemical analysis was performed on frozen sections using a specific polyclonal antibody against Kir7.1, and a monoclonal antibody against Na+–K+ ATPase. Results: Staining with anti–Kir7.1 resulted in punctate labeling of the basolateral membrane of non–pigmented ciliary epithelial (NPCE) cells. A similar labeling pattern was obtained with anti–Na+–K+ ATPase. No immunoreactivity with either antibody was detected in pigmented ciliary epithelial (PCE) cells. Conclusions: Kir7.1 and Na+–K+ ATPase are co–localized in the basolateral membrane of the NPCE cells, where they may function cooperatively to regulate the secretion of aqueous humor.
