Abstract

Abstract: : Purpose: To present three cases of nevus of Ota in a hispanic population and their relation to choroidal melanoma, with histological and ultrastructural analysis. Methods: Three enucleated eyes were evaluated with hematoxylin–eosin and Masson trichrome staining, immunohistochemistry (Prot S100, HMB 45, Melan A)and transmission electron microscopy. The cytologic differences in the transition area between melanocytosis and melanoma were analyzed. Results:Melanocytes in melanocytosis had small oval body and a protruding nucleus with nucleoli and numerous cytoplasmic prolongations. The cytoplasm had moderate number of mitochondrias, smooth and rough endoplasmic reticulum and free ribosomes. Melanine granules were identified within the cytoplasm at several stages of development, including premelanosomes, melanosomes and myelinized granules with an approximate 0.05 micron diameter. Atypical cells showed an altered nuclear morphology with membrane irregularities and lack of organellas. Conclusions: Our case series showed atypical sectors of melanoma within typical ones (melanocytosis). We have not found any previous electron microscopy report of choroidal melanoma in nevus of Ota in the hispanic population.
