Purpose:To examine the rescue effect of cell transplantation with somatic neurotrophic factor gene transfection against constant light damage and to compare the results with the systemic reaction. Methods: We performed somatic transfection of IRES-vector harboring enhanced green fluorescein protein (EGFP) gene or Topo-vector (no EGFP) into rat retinal pigment epithelium (RPE) with or without Axokine, BDNF, bFGF cDNA. MHC-I and -II expression after each gene transfection was examined by real-time PCR. Somatic BDNF gene transfected cells (BDNF-IRES) was recovered after subretinal transplantation by FACS Vantage cell sorting system and examined the expression of MHC antigens as well as BDNF gene. We also examined rescue effect of each cell against constant light damage after subretinal transplantation. Further, we examined the serum reaction against transplanted cells and the percentage of CD4/8 of the rats after transplantation using flowcytometer. Results:BDNF gene transfected cell expressed BDNF gene at the subretinal space almost same level as that of cells cultured on plates by real-time PCR. A statistically significant photoreceptor rescue against light damage was observed at the transplanted sphere when compared to those of vehicle injection and no treatment. Conversely, it was observed only in BDNF at the untransplanted sphere. We confirmed no ectopic migration of the transplanted cells by EGFP. A statistically significant antibody production was observed in Axokine when compared to those of other transplantation. MHC-I gene expression and the percentage of CD8 of Axokine were also higher than those of other cell transplantation. Conclusions: Somatic BDNF gene transfected cell transplantation rescued photoreceptor cell from constant light damage beyond the transplanted area without induction of systemic immunity. Conversely, somatic Axokine gene transfected cell transplantation showed no effect other than the transplanted area and had possibility to induce systemic inflammatory reaction.
