Abstract

Abstract: : Purpose: To investigate the capacity of the PPAR gamma agonist 15 prostaglandin J2 (15 PGJ2) to modulate human RPE cell inflammatory responses Methods: PPAR gamma expression on RPE cells was tested by western blot. Incubation of cells with 15 PGJ2 was immediately followed by stimulation with IFN gamma or TNF alpha. CD54 and MHC class II molecules expression was assessed 48 hours later by flow cytometry. Apoptosis was measured by annexine/PI staining. IL-1 mRNA expression studied by semi-quantitative RT-PCR Results: We showed that ARPE cells and RPE cells express PPAR gamma. 15 PGJ2 down-regulates IFN gamma induced expression of MHC class II molecules, as well as the IFN gamma up-regulation of CD54 on ARPE cells line. At 10uM, 15 PGJ2 had no effects on cell apoptosis. At higher doses, 15 PGJ2 induced cell apoptosis. IL-1 mRNA expression induced by TNF alpha was also inhibited by 15 PGJ2. Conclusions: RPE cells express PPAR gamma in vitro. PPAR gamma ligand 15 PGJ2 modulates pro-inflammatory molecule expression on those cells.
