Abstract

Abstract: : Purpose:To produce monoclonal antibody directed against tumor antigen of retinoblastoma-derived tissue culture cell line SO-Rb50 and purify the SO-Rb50 cells antigen corresponding to the monoclonal antibody of anti-restinoblastoma. Methods: Hybridization was performed and the specificity of the antibody was tested by immunofluorescent and immunohistochemical methods. The purification of the antigen was made using ion-exchange chromatography, DOT-BLOT and SDS-PAGE. Results:Two hybridomas secreted specific monoclonal antibody against retinoblastoma cells were produced,and the isotype of the antibody was IgG2a. After ion-exchange chromatography of SO-Rb50 cells antigen, SDS PAGE only showed a special 83kd unmixed band, stained by the antibody against retinoblastoma cell using WEST-BLOT. Conclusions: The monoclonal antibody is specific and highly active against retinoblastoma cells. The character of the antigen differ from Rb protein of Rb gene product.
