Abstract

Abstract: : Purpose: To investigate the molecular basis for cell growth inhibition or apoptosis in human RPE cells after low-dose radiation. Methods: Cell survival assay, TdT-mediated dUTP-biotin nick-end labeling (TUNEL) assay were performed on post-radiated RPE cell cultures at different periods. Levels of p53, p21, Bax, Fas-FasL were evaluated by semiquantitative reverse transcriptional polymerase chain reaction (RT-PCR). p53 was analyzed by PCR single-strand conformation polymorphism (PCR-SSCP) mutational assay and Western blot. Results: ARPE-19 cell viability decreased with increasing doses of radiation. Apoptosis was detected after 6 hours of radiation. Increase in p53, p21 and Bax mRNA expression by semiquantitative RT-PCR analysis was greatest in the 2-hour post-radiation samples. Transcription levels of Fas and FasL remained unchanged. Protein levels of p53 were increased at the 4- and 6-hour post-radiation samples. Conclusions: Low-dose radiation induces a p53-dependent apoptotic mechanism in RPE cells in vitro.
