Abstract

Abstract: : Purpose: to determine whether glial cell lined-derived neurotrophic factor ( GDNF ) enhances retinal ganglion cell survival in rats with optic nerve transection injury model. Methods: transfection of rAAV-GDNF to ganglion cells was achived by intravitreal injection in adult albino Lewis rats for 3 weeks. The optic nerves were transected intraorbitally about 2 mm from the eye . Two weeks later the neuronal loss was determined by retinal ganglion cell counts ( RGCCs ) through FITC-Dexan back-labeling. Results: RGCCs in AAV-GDNF-treated retina was 380.4cells/mm2 , RGCCs in AAV-lacZ -treated retina was 205.2 cells/mm2 . Conclusion: The preliminary result showed AAV-GDNF exerts an RGC rescue rate of about 40% at 14-days post-axotomy
