Abstract

Abstract: : Purpose: To determine whether the α-amino-3-hydroxy-5-methylisoxazole-4-propionate (AMPA) receptor subunit proteins are expressed in cultured retinal ganglion cells (RGCs). Methods: RGCs were purified from dissociated rat retinal cells (postnatal days 6-8), using a modified two-step panning method and cultured in serum-free medium containing neurotrophic factors and forskolin. Immunohistochemistry was performed on cultured RGCs between one day in vitro (DIV) and 7 DIV using specific antibodies against AMPA subunits GluR1 to 4 and microtubule-associated protein 2 (MAP2), which is essential for dendritic growth by selectively stabilizing dendritic microtubules. Results: The neurites of purified RGCs gradually became elongated in our culture medium. MAP2 protein was expressed in the somatodendritic area of the RGCs from 1 DIV on. GluR1 to 4 proteins were faintly expressed in the somata of RGCs on 1 DIV. They were gradually expressed more strongly compared with 1 DIV in the somatodendritic area on 7 DIV. Conclusions: All AMPA receptors are expressed in the somatodendritic area of cultured RGCs with dendritic outgrowth.
