Abstract: : Purpose: To compare cellular growth and expression of matrix metalloproteases MMP-2 and MMP-9 by cultured epithelial corneal cells on treated amniotic membrane. Methods: Acrylamid gel zymography were performed to compare semi quantitatively MMP's expression by cultured corneal epithelial cells on differents specimen of amniotic membrane. Human limbal explants were cultured in vitro on amniotic membrane using airlifting technique. Amniotic membrane was chemically pre-treated to scratch epithelial monolayer. The culture on acellular amniotic membrane was compared with culture on cellular amniotic membrane. Results: Expanded corneal epithelial cells on acellular membrane formed a confluent epithelium layer in 2 weeks. Epithelial outgrowth was more rapid to confluence on acellular than cellular membrane (p<0.05). Zymography showed MMP's overexpression in cellular amniotic membrane than acellular amniotic membrane when epithelial cells were not a confluent layer. Conclusion: These results correlate the MMP's expression with the cultured epithelium corneal outgrowth on amniotic membrane to support that corneal epithelial cells can be cultured more easily to confluence on acellular amniotic membrane.
