Purpose: To determine genome type of adenovirus (Ad) serotype 8 isolated from the cases of three hospitals with nosocomial infections of epidemic keratoconjunctivitis. Methods: Four strains of Ads, one isolated at September 1998, two isolated at June 2000, and one isolated at August 2001 were analyzed based on a combination of PCR and restriction endonuclease (RE) digestion (PCR-RFLP). Serological analyses were performed for various serotypes of Ad serotype 1 through 8, 11, 19, and 37. Results: 1. The four clinical isolates were neutralized weakly with the antiserum against Ad8 but were not neutralized with other antisera used. 2. The cleavage patterns of 956-bp amprimer of partial hexon, with Hae III, EcoT14 I, Hinf I, Kpn I, Mbo I, and Cfr 13 I, showed the identical cleavage patterns for all isolates. The cleavage patterns of all isolates, with EcoT14 I, Hinf I, Kpn I, Mbo I, and Cfr 13 I, showed identical cleavage patterns to those of Ad8 prototype strain (Trim). However, the cleavage patterns of the isolates, with HaeIII, were not an identical pattern to that of Ad8. The phylogenetic analysis of four isolates and 47 prototype strains based on 956-bp amprimer of partial hexon supported these cleavage patterns with six REs. Conclusion: The findings above indicate that possible new genome type of Ad8 (or new type of Ad subgenus D) was found from nosocomial infections in two hospitals in Japan. Such changes seemed to occur at least in or before1998.
