SPARC (secreted protein, acidic and rich in cysteine) is a matricellular protein known to be integral in extracellular matrix (ECM) assembly. We hypothesize that SPARC may be involved in the development of ocular structures. In this study, we aim to characterize anterior chamber angle parameters in SPARC-null mice.
In this pilot study, eyes of 18 SV129 wild-type (WT) and 9 SPARC-null mice were imaged using the RTVue (Optovue Inc, Fremont, California, USA) anterior segment optical coherence tomography (ASOCT). For all measurements, intraperitoneal injection of ketamine:xylazine were used to anaesthetize the mice. Anterior chamber depth (ACD), angle opening distance (AOD) and the trabecular-iris space area (TISA) were measured in both eyes. Both eyes were analyzed using linear regression analysis and adjusted for age, gender and pupil diameter.
In both left and right eyes, SPARC-null mice had larger TISA 500 AND AOD (p<0.05 for all). After adjusting for age, gender and pupil diameter, SPARC-null mice had deeper ACD (Right [R] β=105.17, p=0.007; Left [L] β=109.29, p=0.01), greater TISA (R β=0.025, p=0.002; L β=0.035, p=0.002) and larger AOD (R β=106.05, p=0.001; L β=145.38, p=0.002) in both eyes. Linear mixed model estimated means of fixed effects also revealed a deeper ACD [mean difference (MD) 145.6μm, standard error (SE) 39.8, p<0.001], greater TISA 500 (MD 0.016mm2, SE.01, p=0.039) and AOD (MD 69.9μm, SE29.9, p=0.028) in SPARC-null mice.
Compared to WT, SPARC-null mice have deeper anterior chamber as well as deeper and wider drainage angles. SPARC is important for the spatial organization of the anterior chamber angle.
