Recently, we reported that hsa-miR-148a-3p (miR-148) was expressed specifically in the vitreous from eyes with rhegmatogenous retinal detachment (RRD) and that miR-148 expression correlated with the size of the retinal breaks and duration of RD. The purpose of this study was to examine correlations between miR-148 expression and inflammatory cytokines from eyes with RRD.
Expression levels of miR-148 and inflammatory cytokines (FGF-2, IFN-γ, IL-10, IL-6, IL-8, MCP-1, TNF-α, and VEGF) were measured in the vitreous fluid of 20 eyes with RRD. Spearman's rank correlation coefficient was used to assess the association between expression levels of miR-148 and the inflammatory cytokines.
The expression levels of miR-148, FGF-2, IL-6, IL-8, MCP-1, TNF-α, and VEGF were 10.8 ± 17.7 folds (relative expression), 22.2 ± 22.6 pg/mL, 90.9 ± 88.4 pg/mL, 42.9 ± 50.7 pg/mL, 7529.8 ± 4828.4 pg/mL, 1.6 ± 4.8 pg/mL, 136.1 ± 128.9 pg/mL, respectively. IFN-γ and IL-10 were under the detection limits in all cases.The results of spearman's rank correlation coefficient were as follows; FGF-2 : r = 0.48, p = 0.03, IL-6 : r = 0.27, p = 0.24, IL-8 : r = 0.44, p = 0.05, MCP-1 : r = 0.08, p = 0.73, TNF-α : r = 0.30, p = 0.20, VEGF : r = 0.18, p = 0.44.
MiR-148 expression in eyes with RRD was significantly correlated with FGF-2, but not with IFN-γ, IL-10, IL-6, IL-8, MCP-1, TNF-α, and VEGF.
