To compare the effects of either 0.2% benzalkonium chloride (BAK) or NaOH on injury-induced sex-specific contributions to increases in pro-inflammatory cytokine mRNA levels associated with: a) corneal wound healing: b) the lacrimal functional unit (LFU), including lacrimal gland (LG) and trigeminal ganglion (TG) in wildtype (WT) and TRPV1-/- mice.
Wound healing responses induced by either 0.2% BAK or 1N NaOH alkali burn (AB) were compared by clinical exams, histology and proinflammatory cytokine gene expression patterns in the cornea, lacrimal gland (LG) and trigeminal ganglion (TG) in male and female C57BL/6 mice (WT) and their TRPV1-/- counterparts after 7 days.
Both BAK and AB corneal injury caused IL-1β, IL-6, and TNF-α mRNA levels to rise by the 7th day in the cornea, and also in LG as well as TG tissues of WT and TRPV1-/- mice. These rises in the pro-inflammatory cytokines were strikingly larger after AB in the cornea, and more evident in TG of WT females than in males. However, AB-induced rises in pro-inflammatory cytokine mRNA levels were larger in the male LG, and TRPV1-/- mice than those in their female counterpart.
Despite the absence of TRPV1 expression, either AB or BAK treatment still induced signature specific increases in corneal, LG, and TG mRNA pro-inflammatory cytokine expression levels. The patterns occurring in males were distinguishable from those in their female counterpart. Therefore, other sex specific receptor-linked signaling pathways besides those linked to TRPV1 contribute to triggering and modulating corneal and LFU responses to injuries.
