Abstract
Purpose :
Congenital cataract is the primary cause of treatable childhood blindness. The morphological features of congenital cataract was complicated. We found the special morphology in congenital cataract with posterior capsule defect (PCD), Furthmore, some patients accompany with sign of uveitis, which indicated the inflammation status in PCD. However, the exact mechanisms of PCD are still poorly understood. The research was to evaluate cytokine profile in aqueous humor of congenital cataract with PCD to search the pathogeny of PCD.
Methods :
We followed a cohort of 30 patients (45 eyes) with congenital cataract from Wenzhou medical university, 18 eyes with PCD and 27 eye with normal congenital cataract as control. Aqueous humor (100–200 μl) was aspirated from each patient by means of limbic paracentesis with the use of 30 gauge needle before the surgery. The procedure was performed under a surgical microscope. The samples were snap frozen and maintained at −80°C until use. Levels of various inflammation-related cytokines in the aqueous humor of using multiplex immunoassay.
Results :
The mean concentration of TGFβ2 (2484.94±1175.7pg/mL) was significantly higher in the PCD group compared to the controls (1108.7±728.2 pg/mL; P =0.004) and the mean concentration of IL-6 (69.3±27.79 pg/mL) was significantly higher in the PCD group than in the controls (2.20±5.75pg/mL; P=0.000). Furthermore, the VEGF, MCP1, IP10 and IL2 were also higher in PCD group. There were no differences in the concentration of TNF-α, IL-1β, PDGFBB, IL-10, IFNγ, IL1α, MIP1α, MIP1β, IL-4, IL-17a, IL-5, GMCSF, Eotaxin. In PCD group, 7eyes complied with uveitis, while the cytokines were not significant compared with PCD without uveitis.
Conclusions :
Higher preoperative levels of TGFβ2, IL-6, VEGF in aqueous humor were associated with posterior capsule defect, which indicated the inflammation state in congenital cataract with posterior capsule defect.
