Intravitreal bromfenac with liposomes. A toxicology study in rabbit eyes.

Abstract

Purpose : NSAIDs (Nonsteroidal Anti-Inflammatory Drugs) have demonstrated to have an important role in the treatment of cystoid macular edema and have a potential effect on diabetic macular edema.
The purpose of the present study was to evaluate the toxicity of intravitreal liposomes with bromfenac in rabbit eyes.

Methods : We evaluated the toxicity in rabbit eyes (n= 10 cases and n=10 controls) after intravitreal bromfenac (450 ng/ 0.1 ml) in combination with liposomes. All animals had pre-treatment and at the end of treatment (6 weeks) comprehensive eye examination, OCT and ERG. Histopathology exam was also performed in all eyes.

Results : All eyes, control and studied showed at the beginning of the study and at the end normal eye examination. Anterior segment, vitreous, retina, and optic nerve did not show abnormalities or signs of toxicity. Pretreatment and post-treatment OCT and ERG were normal in all studied eyes. Histopathology examination in either control and treated were normal without evidence of toxicity.

Conclusions : Intravitreal liposomes with bromfenac at 450 ng/ 0.1 ml dose are not toxic in rabbit eyes. Our results warrant further studies in human eyes.
