Abstract

Purpose : to assess the interocular variation and the correlation between structural and functional parameters by using optical coherence tomography (OCT) and microperimetry in subjects with Usher syndrome-associated retinitis pigmentosa (RP).

Methods : in this cross-sectional study, best corrected visual acuity (BCVA) and contrast sensitivity (CS) were assessed with ETDRS and Pelli-Robson charts respectively; mean retinal sensitivity (MRS) in the central 10 degrees was obtained using a customized 27-point grid. An OCT macular scan 20°x20° wide (97 horizontal B-scans) was centered to the fovea to obtain the macular volume (MV) and the central retinal thickness (CRT). The agreement between eyes was tested with the Bland-Altman method and here presented as relative interocular difference (RID) values (mean differences expressed as percentage of the mean values of all eyes). Correlations between age, structural and functional parameters were calculated using Generalized Estimating Equation.

Results : forty eyes of 20 patients (8 females, age: 38 ± 17.4 years [range: 15-72 years]) were included. Symmetry between eyes was demonstrated for all considered parameters: MV and CRT showed RID values of 2,3% and 1%; functional data had RID values of 4.2%, 3.4%, and 2.1% (MRS, BCVA, and CS respectively). No significant correlations were found between structural and functional parameters. However, age was significantly correlated with BCVA (coefficient [B] = 0.15, p=0.01), CS (B=-0.17, p<0.01), and MRS (B = -0.15 p<0.01), but not with OCT values.

Conclusions : in general, good interocular agreement exists in this cohort of patients with syndromic RP. Although no correlations were found between structural and functional parameters, functional decline seems to be evident with increasing age (similar to previous studies on non-syndromic RP). Further investigations with prospective data are needed to confirm these findings.
