Abstract

Purpose : We previously reported retinal gliosis and microglia distribution in rat retina at 8 months following ABO exposure. Here, we examined changes in intermediate filament assembly and composition at 48 h and 1 wk post-ABO exposure and performed serum cytokine profiling to assess ABO-associated serum biomarkers.

Methods : Male Long-Evans rats were anesthetized and given a single ABO exposure (see Allen RS et al, J. Neurotrauma, 2018); non-exposed age/sex-matched rats served as controls. Tissues and serum were harvested at 48 h and 1 wk post-ABO exposure. Cryosections were prepared from fixed whole eyes oriented and embedded in OCT. Immunohistochemistry was performed using antibodies against GFAP (Müller glia), nestin, and peptidyl citrulline, followed by detection with fluor-conjugated secondary antibodies and confocal microscopy. Cytokine profiling was performed on sera collected at 48 h post-ABO using a Proteome Profiler® Rat Cytokine Array (R&D Systems), imaged and quantified with a Bio-Rad ChemiDoc™ System. Statistical significance: P<0.05 (Student’s t-test), N=6/group.

Results : Ipsilateral neural retinas (I-ABO) exhibited significantly elevated GFAP levels and reactive gliosis, vs. controls, at 48 h and 1 wk post-ABO exposure; however, no differences in optic nerve head GFAP labeling were observed (I-ABO vs. control). Surprisingly, citrullination of filaments was comparable in I-ABO and control retinas. Nestin immunoreactivity in neural retinas exhibited a fine filamentous or diffuse appearance in controls, as well as some endothelial labeling. Nestin immunolabeling was significantly increased in I-ABO vs. control retinas (with regional variability) at 48 h, in a radial pattern similar to that of GFAP, but was less elevated at 1 wk vs. 48 h post-ABO. Serum cytokine profiling revealed significantly elevated levels of TIMP-1 and CXCL7 at 48 h post-ABO exposure, vs. controls.

Conclusions : Discrete alterations in retinal intermediate filament composition and assembly with attendant reactive gliosis occur at early time points (48 h and 1 wk) post-ABO exposure. TIMP-1 and CXCL7 may provide useful serum biomarkers to correlate with ABO-induced retinal injury.
