Chronically activated unfolded protein response (UPR) in mice with retinal degeneration may launch a pro-apoptotic program associated with oxidative stress, pro-inflammatory signaling, dysfunctional autophagy, free cytosolic Ca2+ overload, and altered protein synthesis rate. The research focuses on the study of regulatory mechanisms of UPR activation and offers a translational platform to overcome molecular consequences of retinal pathogenesis. The role of the UPR PERK arm and its downstream targets will be discussed to highlight the therapeutic benefits of reprogramming PERK signaling in preclinical animal models of retinal degeneration.
