Abstract

Purpose : Recent studies have reported the anti-inflammatory effect of Lithospermum erythrorhizon. We investigated the effect of ethanol extract of Lithospermum erythrorhizon (EELE) on endotoxin-induced uveitis (EIU) in rats and mice. In addition, the endotoxin-induced expression of pro-inflammatory cytokines and activation of transcription factor IRF and NFκB were investigated in human monocyte reporter cell lines (THP1) treated with EELE in vitro, to clarify the anti-inflammatory effect.

Methods : Endotoxin-induced uveitis was induced in rat and mice by a footpad injection of lipopolysaccharide (LPS). EELE (10 mg/kg) was daily administered for 7 days before LPS injection. After 24 hours of LPS injection, clinical severity and retinal vessel thickness were evaluated. THP1-Blue-ISG and THP1-XBlue cells pretreated with EELE were incubated with LPS for 24 hours. Level of TNFα, IL-6 and IL-8 expression and activation of IRF and NF-κB were investigated.

Results : Clinical severity and retinal vessel thickness were significantly decreased in EELE-treated rat than in control group (P < 0.001, P = 0.0167). EELE treatment significantly reduced the levels of infiltration cells, protein, and PGE2 in the aqueous humor. EELE down-regulated the production of TNFα, IL-6 and IL-8 in THP1 cells. And EELE treatment suppressed the activation of NFκB/AP1 and IRF.

Conclusions : Orally administrated EELE alleviated the ocular inflammation. These findings demonstrate that EELE could be an effective agent for the control of endogenous ocular inflammatory disease.
