Abstract

Purpose : We have reported that violet light (VL, 360–400 nm wavelength) is effective in suppressing myopia progression in the chick and mouse models (Torii et al, EBioMedicine. 2017, Jiang X et al. PNAS. 2021) and that the expression level of early growth response-1 (EGR1), myopia-suppressive gene in the retina was increased by VL exposure. The myopic inhibitory effect of VL was reduced in retina-specific OPN5 knockout (KO) mice. However, the upregulation of EGR1 expression by VL remains unclear. In this study, we examined whether the upregulation of EGR1 by VL is dependent on OPN5 in vitro and in vivo experiments.

Methods : 661w, murine retinal ganglion precursor-like cell line and OPN5 KO cells by CRISPR gRNA were exposed to VL (250 µW/cm2). Retina specific OPN5 KO mice (Opn5 flox/flox; Chx10-cre+/-) by mating Opn5 flox mice and Chx10-cre mice and their littermate controls (Opn5 flox/flox; Chx10-cre-/-) were exposed to VL (400 µW/cm2). The level of EGR1 gene and protein expression in cells and mouse retina were analyzed using real time-PCR and western blotting.

Results : EGR1 gene expression level in 661w cells increased after 3 hours of VL exposure and was higher than that in OPN5 KO cells (3.46±1.96 vs. 1.04±0.52, two-tailed Student’s t-test, p=0.015). We also confirmed the increase in EGR1 protein expression of 661w cells by VL exposure. After 7 hours of VL exposure to the mice, the control group increased in EGR1 gene expression of the retina and was higher than that in retina specific OPN5 KO mice (3.47±0.67 vs. 2.36±0.25, two-tailed Student’s t-test, p=0.021).

Conclusions : We showed that EGR1 expression in the retina was upregulated by VL mediated by OPN5 in vitro and in vivo experiments.
