In order to achieve vision restoration for optic neuropathies via retinal ganglion cell replacement, we need a clear roadmap as to how to promote donor ganglion cell rewiring with existing retinal circuitry in a diseased eye. A sound understanding of inner retinal wiring and the ability to establish connectivity with appropriate synaptic partners are central to the anatomical and functional success of cellular approaches for retinal regeneration. This talk will discuss our current understanding as well as unanswered questions including how best to maintain synaptic and dendritic integrity, the importance of glial and innate immune responses on inner retinal circuitry, and the identification of cell-intrinsic and cell-extrinsic cues that underlie inner retina circuit development and leveraging this information to promote donor ganglion cell integration into degenerating retinal circuits.
