By performing non-invasive time-lapse imaging of the cornea in healthy humans, we identify that T cells make up a substantial portion of cells in the epithelium, which have been misclassified as dendritic cells using static imaging. Our data establish the utility of Fun-IVCM as a tool to identify concurrent alterations to the static and dynamic features of T cells, DCs, and macrophages, across the depth of the tissue. How to apply this technique and what is the key to successfully acquire high-quality time-lapsed videos will be presented. Factor of season and factor of age on the behaviour of corneal immune cell subset is also examined.
